چکیده

PURPOSE The retinal blood flow is regulated by the tone of resistance arterioles, which is influenced by purinergic compounds such as adenosine and adenosine 5'-triphosphate (ATP) released from the retinal tissue. However, it is unknown what cellular elements in the perivascular retina are responsible for the effect of purines on the tone of retinal arterioles. METHODS Porcine retinal arterioles were loaded with the calcium-sensitive fluorophore Oregon green. The vessels were mounted in a confocal myograph for simultaneous recordings of tone and calcium activity in cells of the vascular wall during stimulation with ATP and adenosine, with and without modifiers of these compounds. Additionally, immunohistochemistry was used to localize elements with calcium activity in the vascular wall. RESULTS Hyperfluorescence indicating calcium activity was recorded in a population of abundant round boutons interspersed in a network of vimentin-positive processes located immediately external to the smooth muscle cell layer but internal to the perivascular glial cells. These structures showed calcium activity when the vessel was relaxed with ATP but not when it was relaxed with adenosine. Ryanodine reduced calcium activity in the boutons, whereas the ATP antagonist adenosine-5'-O-(α, β- methylene diphosphate) reduced calcium activity in both the boutons and vascular tone. CONCLUSIONS The vasodilating effect of purines in porcine retinal tissue involves ATP-dependent calcium activity in a layer of cellular boutons located external to the vascular smooth muscle cells and internal to the perivascular glial cells.

Blood Flow Autoregulation in the Retina

Autoregulation of blood flow, the maintenance of relatively constant blood flow despite variations in pressure, is characteristic of many tissues, including the retina. Impaired retinal autoregulation has been shown to be a risk factor for glaucoma, suggesting that the relation between vascular regulatory mechanisms and glaucoma progression should be investigated. Here, a vascular wall mechanic...

N-methyl-D-aspartic acid causes relaxation of porcine retinal arterioles through an adenosine receptor-dependent mechanism.

PURPOSE Disturbances in retinal perfusion due to impaired regulation of vascular tone are believed to be involved in the pathogenesis of several vision-threatening retinal diseases. Two recent studies have shown that the glutamate receptor agonist, N-methyl-D-aspartic acid (NMDA), and adenosine induce relaxation of isolated porcine retinal arterioles in vitro. However, it remains to be elucidat...

Purinergic control of vascular tone in the retina.

Purinergic control of vascular tone in the CNS has been largely unexplored. This study examines the contribution of endogenous extracellular ATP, acting on vascular smooth muscle cells, in controlling vascular tone in the in vivo rat retina. Retinal vessels were labelled by i.v. injection of a fluorescent dye and imaged with scanning laser confocal microscopy. The diameters of primary arteriole...

Resveratrol, a component of red wine, elicits dilation of isolated porcine retinal arterioles: role of nitric oxide and potassium channels.

PURPOSE Resveratrol, a polyphenolic phytoalexin found in grapes and red wine, has been shown to exert cardiovascular benefits, but its action in the retinal microcirculation remains unknown. In this study, the direct effect and the underlying mechanism of the vasomotor action of resveratrol were examined in retinal arterioles. METHODS Porcine retinal arterioles were isolated, cannulated, and ...

Wall-to-lumen ratio of retinal arterioles as a tool to assess vascular changes.

The retina offers a beautiful and unique opportunity to visualize and examine the body’s microvasculature safely, repeatedly, quickly, and noninvasively in vivo. Retinal arterioles appear to undergo similar changes as cerebral and peripheral arterioles in hypertension, indicating that retinal arteriolar abnormalities mirror structural and functional microvascular changes elsewhere in end-organ ...
